Caracterización de los polimorfismos del gen PRNP en ovinos Zom
Resumen
Objetivo del estudio: Los polimorfismos en los codones 136, 154 y 171 del gen de la proteína priónica (PRNP) son fundamentales debido a su asociación con la susceptibilidad o resistencia al scrapie, una enfer-medad neurodegenerativa mortal en ovinos. El objetivo de este estudio fue analizar la diversidad genética del PRNP en ovinos Zom, una raza autóctona de Turquía, y evaluar sus posibles implicaciones en relación con la resistencia frente al scrapie.
Área de estudio: Turquía.
Material y métodos: A partir de muestras de sangre obtenidas de 46 ovinos Zom. Se amplificó una región de 662 pares de bases del gen PRNP mediante la reacción en cadena de la polimerasa (PCR) y posteriormente se procedió a su secuenciación. Se calcularon las frecuencias alélicas y genotípicas de los polimorfismos identificados. Los posibles efectos funcionales de los polimorfismos de nucleótido único (SNPs) de tipo mis-sense fueron predichos utilizando herramientas bioinformáticas como PolyPhen-2, PANTHER y AMYCO. Asimismo, se llevó a cabo un análisis de haplotipos con el fin de evaluar la variabilidad genética.
Resultados principales: Se identificaron cinco alelos y seis genotipos. El alelo ARQ y los genotipos ARQ/ ARQ y ARR/ARQ fueron predominantes, mientras que los alelos y genotipos asociados con la resistencia fueron menos frecuentes. Se identificaron diez polimorfismos, entre ellos una variante de inserción/deleción y seis SNPs de tipo missense (mutación de sentido erróneo) que originaron sustituciones de aminoácidos. El análisis con PolyPhen-2 clasificó las variantes T195I, T196S y N146S como benignas, mientras que T193S se predijo como posiblemente perjudicial y V179G como probablemente perjudicial. PANTHER predijo que todos los SNPs evaluables podrían tener efectos perjudiciales, en tanto que AMYCO no identificó riesgo amiloidogénico. Además, se identificaron doce haplotipos distintos dentro de la población analizada.
Aspectos destacados de la investigación: Este estudio proporciona datos a nivel poblacional sobre los poli-morfismos de PRNP en ovinos Zom, lo que sugiere un vínculo potencial con la resistencia parcial al scrapie y ofrece información para futuros enfoques de reproducción.
